Etiopathogenesis of cervical cancer

Lucia Kocová, Oliver Sadovský, Tibor Lengyel

Persistent infection with high-risk human papillomavirus, which is associated with integration of viral genome into the host cell chromosome has an essential role in cervical cancer pathogenesis. Carcinogenesis is a consequence of overexpression of viral oncogenes, genome instability and deregulated cell proliferation. In malignant transformation, there play a key role viral oncoproteins E6 and E7 that interact with proteins important in the cell cycle regulation. For a long time, cervical precanceroses and invasive cancers have been considered to originate from HPV infection of the basal cell layer of the squamous epithelium of the transformation zone or ectocervix. Recent studies uncovered a population of embryonic squamocolumnar junction cells and prove their critical role in the pathogenesis of cervical (pre)cancers.
